Anemone coronaria var. coccinea'da androgenesis çalışmaları için yygun çicek tomurcuğu morfolojisinin tespit edilmesi

Anemon cinsi içerisinde, en fazla kültürü ve ıslah çalışmaları yapılan tür Anemone coronaria L.’dır. Ancak türdeki yüksek heterozigoti nedeniyle, saf hatlara ulaşma süreci en az 8-10 yıl gerektirmektedir. Bu problem ıslahçıları haploidizasyon tekniklerine yöneltmiş ve şimdiye kadar yürütülen az sayıdaki haploidizasyon çalıĢmalarında androgenesis yöntemleri tercih edilmiştir. Ancak, bu çalışmalarda kullanılabilecek uygun tomurcuk morfolojisine ilişkin ayrıntılı bilgiye rastlanmamıştır. Çalışmada, doğal A.coronaria var. coccinea tomurcukları içerisindeki mikrosporların geliĢim aĢamalarını belirlemek ve bunlara göre tek çekirdekli mikrosporlara sahip uygun çiçek tomurcuğunun tanımını yapabilmek amacıyla, farklı büyüklükteki tomurcuklar morfolojilerine göre 10 gruba ayrılmış ve bu tomurcuklar içerisindeki mikrosporların gelişim aĢamalarını belirlemek için farklı sitolojik ve histolojik boyama yöntemleri kullanılmıĢtır. Bulgulara göre, A.coronaria var. coccinea androgenesis çalışmaları için en uygun safhalar oldukları kabul edilen geç tetrat ve tek çekirdekli safhadaki mikrosporları, genellikle 2 ve 3 numaralı tomurcukların içerdikleri belirlenmiĢtir. Bu tomurcukların makroskobik göstergeleri ise; genç tomurcukların özellikle toprak yüzeyinde yeni gözükmeye baĢladığı sıralarda eğik durması ve çiçek sapıyla neredeyse paralele yakın Ģekilde oldukça dar bir açı oluşturması, ortalama 6x9 mm büyüklükte olmaları ve involukrum yaprakların henüz açılmaması nedeniyle tepallerin dıĢarıdan görünmemesi olarak tanımlanmıĢtır. ÇalıĢmada ayrıca, bir tomurcuk içerisindeki anterlerin asenkronik gelişim biçimi gösterdiği, bir anter içerisindeki mikrosporların ise homojen Ģekilde geliĢtikleri tespit edilmiştir.

Determınatıon of suıtable flower bud morphology for androgenesıs studıes in Anemone cornaria var. coccinea

The most intensive cultivation and breeding studies conducted in the genus Anemone has been actualized in Anemone coronaria L. species. However, the inbreeding process takes at least 8-10 years because of high level of heterozygosity in the species. The trouble in the obtaining uniform pure lines has oriented the breeders towards the haploidization techniques. In few haploidization studies performed in A.coronaria so far, anther culture technique was preferred. But, it could not be found detailed literature about the morphology of suitable flower bud to be used in androgenesis studies. In the study, it was aimed to determine the developmental stages of microspores in the flower buds of wild A.coronaria var. coccinea and to describe the morphology of the suitable bud having uninucleate microspores. For this purpose, the flower buds in different size were separated into 10 groups. Different cytologic and histological dying techniques were used for identification of the microspores’ developmental stages. According to the findings, the flower buds in the second and third group containing the microspores in the stages of late-tetrad and uninucleate were determined to be the most suitable buds for androgenesis studies of A.coronaria var. coccinea. The morphological markers of them have been described as: Young buds stands curved especially when they first appears on the soil surface and creates a quite narrow angle almost parallel with the pedicels. They are approximately 6x9 mm in size and their tepals do not seen from the outside since the involucrum leaves are closed. Additionally, it was determined that the microspores in an anther are homogeneously developed while the anthers within a bud show asynchronous development.
