Sığır vebası virüsü rekombinant hemaglutinin proteinin enzim immunoassayda kullanımı

Bu çalışmada, Staphylococcus aureus'un protein A bölgesiyle füzyon edilmiş şekilde Escherichia colîĞe açıklatılan ve IgG affiniti kromatografisiyle saflaştırılan sığır vebası virüsünün (rinderpest virus; RPV) rekombinant hemaglutinin (rH) proteininin enzim immunoassayda (EIA) kullanımı araştırılıştır. Bu amaçla, rH proteininin N'-ucunda bulunan protein A'ya özgül olmayan IgG bağlanmalarını engellemek için, rH bağlı EIA kapları önce insan serumuyla bloklanmıştır. Daha sonra, rH proteini ve RPV antijeni bağlı EIA kaplarında deneysel immunize fare serumlarına aitabsorbans değerleri kıyaslanmıştır. Çalışmanın sonuçları, çalışmamızda rapor edilen yöntemin stafilokokal protein A bölgesine sahip rH proteininin immünolojik deneylerde güvenilir bir şekilde kullanılma imkanı sağlandığını göstermiştir.

The use of recombinant hemagglutinine protein of rinderpest virus in enzyme immunoasay

In this study, Rinderpest virus (RPV) recombinant hemagglutinine protein (rH) fused with protein A region of Staphylococcus aureus was expressed in Escherichia coli and purified by IgG affinity chromatography. rH protein was also used to establish enzyme immunoassay. Therefore, to prevent IgG binding to the protein A the wells coated with the rH proteins were blocked by human serum. Afterwards, RPV antigens were added to the wells to evaluate this assay. To this end, serum from mice immunized with RPV was used for EIA. Our data indicated that the rH protein fused with protein A of Staphylococcus aureus can be reliably used for enzyme immunoassays.
