Ratlarda deneysel olarak oluşturulan intraabdominal adezyonların önlenmesinde aprotinin ile metilen mavisinin etkinliğinin karşılaştırılması

Bu çalışma; ratlarda deneysel olarak oluşturulan intraabdominal adezyonları önlemede aprotinin ve metilen mavisinin etkinliğini araştırmak amacıyla yapılmıştır. Ağırlıkları 175-200 gr arasında değişen 45 adet erkek rat rastgele 15'erli üç gruba ayrıldı. Ratlar eter ile anesteziye alındı. Steril şartlar altında karın boşluğuna 3 cm'lik bir ensizyonla girildi. Tüm ratlarda karın duvarının iç yüzünde ve sekum serozasında steril bir diş fırçasıyla hasar oluşturuldu. Birinci gruptaki ratlara tek doz 25000 KIU (2.5 mi) aprotinin, ikinci gruptaki ratlara tek doz %1'lik metilen mavisi 2.5'mi, üçüncü gruptaki ratlara ise yine tek doz 2.5 mi serum fizyolojik intraperitonal olarak uygulandı. Karın boşluğu 3/0 ipek iplikle kapatıldı. On beş gün sonra tüm ratlar yüksek dozda eterle ötenazi edilerek laparatomi yapıldı ve adezyonlar 0-3 arasında değişen derecelerle derecelendirildi Birinci gruptaki radarın 13 tanesinde adezyon görülmezken, 1 tanesinde birinci derecede, 1 tanesinde ise ikinci dereceden adezyon gözlendi. İkinci gruptaki ratların 7 tanesinde adezyon görülmezken, 4 tanesinde birinci derecede, 2 tanesinde ikinci derecede ve 2 tanesinde ise üçüncü dereceden adezyon gözlendi. Üçüncü grupta 1 ratta adezyon gözlenmezken, 12 ratta üçüncü derecede ve 2 ratta ikinci derecede adezyon gözlendi. Her iki grup, kontrol grubu ile adezyon açısından karşılaştırıldığında aradaki fark istatistiksel olarak önemli bulundu.

Effects of Aprotinin and Methylene Blue in Prevention of Experimentally Performed intraabdominal Adhesions in Rats This study was aimed to carry out a research on the effects of aprotinin and methylene blue in the prevention of experimentally performed adhesions in the rat models. Forty five male rats, weighed between 175-200 gr were randomly divided into three groups of 15 rats each. After anesthesia with ether and preparation of the surgical field for aseptic surgery an incision of 3 cm in length was performed along the abdominal midline. The serosal layer of the cecum and the inner surface of abdominal wall corresponding to the cecum were abraded aseptically with a tooth-brush. Groups 1, 2 and 3 rats were given intraperitoneally a single dose of 2.5 ml aprotinin (25000 KIU), methylene blue (1 %) and saline; respectively. The abdominal incision was closed with 3/0 silk suture. Fifteen days after the surgery all rats were killed with overdose administration of ether, their abdominal cavities were exposed and the adhesions occurred were graded on a scale of 0-3. In group 1, there were 13 rats with grade 0,1 rat with grade 1 and 1 rat with grade 2 adhesion. In group 2, there were 7 rats with grade.0,4 rats with grade 1, 2 rats with grade 2 and 2 rats with grade 3 adhesion. In the control group there was-1 rat with grade 0, 2 rats with grade 2 and 12 rats with grade 3 adhesion.
