Kedi ve köpek korpus siliyaresinde bağ doku mast hücreleri ve farklı boyanma özellikleri
Bu çalışına kedi ve köpeğin korpus siliyaresinde bağ doku mast hücrelerinin varlığını ve graniil yapılarım ortaya koymak amacıyla yapıldı. Bu amaçla nötiir formolde tespit edilen gözlerden alınan kesitlerin bir bölümü bağ doku mast hücrelerinin varlığını ortaya koymak amacıyla metakromazi için tolııidin blue ile boyandı. Diğer kesitler ise bağ doku ınast hücre granüllerinin farkını ortaya koymak amacıyla methylgreen-pyronin yöntemiyle beyandılar. Tolııidin blue ile boyamada her iki hayvan grubunda korpus siliyarede çok seyrek bağ doku mast hücrelerine rastlandı. Methylgreen-pyronin ile boyama sonucunda kedi korpus siliyaresinde bağ doku ınast hücre granülleri kırmızı renge, çekirdekleri de soluk maviye boyandı. Bunlar ınast hücresine özgü tipik yapıları ile farkedildiler. Köpek korpus siliyaresinde ise bağ doku ınast hücreleri methylgreen-pyronin ile boyanmaclılar. Sonuçta, köpek ve kedi korpus siliyaresinde çok seyrek bağ doku mast hücreleri varlığı ortaya kondu ve kedi bağ doku ınast hücrelerinde granüllerin methylgreen-pyronin ile kırmızıya boyandığı köpeklerinkinin ise boyanmadığı görüldü. Bu nedenle iki hayvan grubunda korpus siliyare bağ doku ınast hücre granül içeriklerinin farklı olabileceği ortaya kondu.
Presence and different staining properties of connective tissue mast cells in corpus ciliare of cat and dog
This study was carried out to investigate the presence and granule structure of connective tissue mast cells in the corpus ciliare of cat and dog. In order to demonstrate the presence of connective tissue mast cells, some of the tissue sections in neutral formalin were stained metachromatically with toluidine blue. Other tissue sections were also stained with methylgreen-pyronin method to demonstrate the different mast cell granules. During staining with toluidine blue, very rare connective tissue mast cells were found in the corpus ciliare of both animal groups. After staining with methylgreen-pyronin, the connective tissue mast cell granules were stained in red and mast cell nucleus in pale blue in the cat corpus ciliare. These were noticed by the typical structure to mast cells. In the dog corpus ciliare, connective tissue mast cells were not stained with methylgreen-pyronin. As a result, presence of very rare connective tissue mast cells in the corpus ciliare of cat and clog were demonstrated. Also, it was observed that the granules in the cat connective tissue mast cells were stained in red by methylgreen-pyronin, in contrast, the granules in the dog connective tissue mast cells were not stained. These results indicated that the granule structure of connective tissue mast cells in the corpus ciliare of these two animal groups could be different.
