Metabolik sendromda paraoksonaz-1 aktivitesi ve koroner arter hastalığı ilişkisi

Paraoksonaz-1 kalsiyum bağımlı antioksidan bir enzimdir. Paraoksonaz-1’in HDL yapısında bulunduğunun anlaşılmasından sonra koroner arter hastalığı ile ilişkisine yönelik çalışmalar artmıştır. Metabolik sendrom kriterlerinden biri olan dislipidemi koroner arter hastalığı için de risk faktörlerinden biridir. Amacımız metabolik sendromlu hastalarda Paraoksonaz-1 aktivitesini belirleyerek, metabolik sendromlu kişilerde koroner arter hastalığı ile ilişkisini araştırmak, paraoksonaz-1 aktivitesinin HDL, trigliserid ve oksidatif stres’in biyokimyasal göstergelerinden malondialdehid ile korelasyonunu göstermektir. Çalışmamızda ‘Uluslararası Diyabet Derneği’ kriterlerine göre metabolik sendrom tanısı alan 100 hasta ve 26 sağlıklı kişinin örnekleri değerlendirildi. Paraoxonase-1 activity in the metabolic syndrome and its relationship with coronary artery disease

Human serum paraoxonase-1 (PON-1) is a calciumdependent antioxidant enzyme. After it was understood that PON-1 is found in the structure of HDL, a number of studies related to its correlation with coronary artery disease (CAD) has increased. One of metabolic syndrome (MS) criteria is dyslipidemia. It is also a risk factor for CAD. Our aim is to evaluate activity of PON-1 in patients with metabolic syndrome, to explain its relationship with HDL, triglyceride (TG) and malondialdehyde (MDA) which is one of the biochemical parameters of oxidative stress. Our study was carried out with 26 healthy subjects and 100 metabolic syndrome patients according to the definition of the International Diabetes Federation. We found significantly lower PON-1 activity and higher MDA levels in the serums of MS patients (p<0.05) compared to healthy subjects. MS patients with CAD have lower levels of PON-1 activity and higher levels of MDA compared to MS patients without CAD (p<0.05). There was not any correlation between PON-1 activities and HDL levels (p>0.05, r=0.15), also between PON-1 and TG levels (p>0.05, r=0.14) of MS patients. As a result; we concluded that PON-1 activity may be used as CAD criteria in MS patients.
