Pirinç Biti Sitophilus oryzae Popülasyonlarında Wolbachia’nın Belirlenmesi

Özet 1 :Sitophilus oryzae (Coleoptera: Curculionidae ) başta mısır olmak üzere tüm hububat çeşitlerinde zararlıdır. Hububat danelerinde beslenmeleri sonucunda üründe kalite ve ağırlık yönünden kayıplar meydana getirir. Endosimbiyont bakteri olan Wolbachia, eklembacaklıların %66’sında tespit edilmiş olup konukçularının üremesi üzerinde partenogenesis, erkek öldürücülük, feminizasyon ve sitoplazmik uyumsuzluk şeklinde üreme bozukluğuna neden olan önemli bir bakteridir. Çalışmada Ege (İzmir, Manisa), İç Anadolu (Ankara, Konya), Karadeniz (Samsun), Güneydoğu Anadolu (Diyarbakır, Şanlıurfa) ve Akdeniz (Adana) bölgelerinden toplam 12 lokasyondan S. oryzae popülasyonları toplanmış ve moleküler yöntemler ile Wolbachia varlığı araştırılmıştır. Çalışmada DNA ekstrasyonunu takiben PCR reaksiyonları wsp primerleriyle taranmıştır. Jel görüntülerine göre, taranan 120 örnekten 118 (%98,33)’inde Wolbachia tespit edilmiştir. Şanlıurfa ve Adana (Çukurköprü)’da %90 oranında, Manisa, İzmir, Ankara (Haymana-Karakeçeli), Diyarbakır, Samsun, Konya (Merkez-Alibeyköy) ve Adana (Ceyhan) illerinde ise %100 oranında Wolbachia varlığı tespit edilmiştir. Bu çalışma ülkemizde S. oryzae üzerinde Wolbachia varlığının belirlendiği ilk çalışmadır.Özet 2 : Sitophilus oryzae (Coleoptera: Curculionidae ) is the harmful insect for grains and especially for corn; as a result of consuming grains, it leads to quality and weight loss in products. Wolbachia, an endosimbiyont bacteria, has been found in %66 of arthropods, and causes various reproduction disorders including partenogenesis, male lethality, feminisation and cytoplasmic incompatibility. In the present study, S. oryzae populations were collected from 12 locations in Agean region (Izmir and Manisa), Central Anatolia region (Ankara and Konya), Black Sea region (Samsun), South-eastern Anatolia region (Diyarbakir and Şanlıurfa) and Mediterranean region (Adana). Existence of Wolbachia was investigated through molecular methods. Specifically the PCR reactions following DNA extraction were scanned with Wsp primers. According to gel displays, Wolbachia was determined in 118 out of 120 samples. In particular, the samples in Şanlıurfa and Adana (Cukurkopru) contained Wolbachia as 90%, and the samples in Manisa, Izmir, Ankara (Haymana-Karakeceli), Diyarbakir, Samsun, Konya (Merkez-Alibeyköy) and Adana (Ceyhan) infected Wolbachia as 100%. The study is the first study on determination of Wolbachia in S. oryzae populations in Turkey.
