[Tylenchida: Meloidogynidae])'na Karşı Dayanıklılık Düzeylerinin Saptanması Üzerine Araştırmalar

Çalışmada, in vitro koşullarda yetiştirilen anaçlardan Garnem, Cadaman ve Myrobalan 29 klonunun kök-ur nematodları Meloidogyne incognita ve M. javanica'ya dayanıklılık düzeyleri araştırılmıştır. Bu amaçla, Garnem, Cadaman genotiplerine ait sürgün uçları, 2.0 mg/l BAP + 0.05 GA3hormonu içeren katılaştırılmış MST ortamında, Myrobalan 29 klonu ise 1.0 mg/l BAP içeren katılaştırılmış MS ortamında kültüre alınmışlardır. Farklı IBA konsantrasyonlarını içeren MS ve MST ortamları sürgünlerin köklendirilmesi amacıyla kullanılmıştır

In Vitro Koşullarda Yetiştirilen Bazı Sert Çekirdekli Meyve anaçlarının Kök-Ur Nematodları (Meloidogyne incognita ve Meloidogyne javanica

In this study, in vitro grown Garnem, Cadaman, and Myrobalan 29C were tested for resistance to root-knot nematodes, M. incognita race 2 and M. javanica. In vitro clonal propagation of Garnem and Cadaman rootstocks were used MST medium with 2.0 mg/l BAP + 0.05 GA3. In vitro clonal propagation of Myrobalan 29 C was used MS medium supplemented with 1.0 mg/l of BAP. MS and MST media suplemented with different concentration of IBA were used for rooting of shoots. Potted plants were inoculated with 1000 second stage of juveniles (J2) of M. incognita race 2 and M. javanica. Three months after inoculation, the ratio of galling in the roots and the number of nematod in the soil of infested rootstocks were recorded. Reproduction ratio of M. incognita race 2 and M. javanica was calculated as 0 for all tested rootstocks. According to evaluation of gall ratio, gall index of Cadaman was 1 and the gall index of the rest of rootstocks was 0.
