Callus Development in Embryo Culture of Wheat

In this study mature and immature embryos of Doğu-88 wheat variety was cultured for callus development on MS medium supplemented with 4 different substances. Experiment was planned and analysised as factorial design with two replication. Data has been converted with rootsquare transformation. Observations were continued during 42 days. Number oftubes in which callus, shoot and root formation occured, were found after observation. The immature embryos formed the highest callus (average 94 % ) on medium MS containing 2 mg/l 2,4-D + 1 mg/l kinetin. The mature embryos formed the highest callus ( average 95 % ) on MS medium containing 2 mg/l 2,4-D. The mature embryos gave the best result with respect to callus weights. The highest callus weights were obtained (average 395.3 mg) on MS medium containing 2 mg/l =2,4-D + 1 mg/l kinetin.
