Erzincan İlinde Fasulye Baklalarından İzole Edilen Rhizoctonia Türlerinin Anastomosis Grupları ve Patojenisitesi

Bu çalışma, Erzincan ilinde fasulye bitkilerinin toprak üstü kısımlarında ağ yanıklığı hastalığına neden olan Rhizoctonia izolatlarının anastomosis gruplarını (AG) ve patojenisitelerini belirlemek amacı ile yapılmıştır. Yapılan izolasyonlar sonucu fasulye baklalarından 38 Rhizoctonia izolatı elde edilmiş olup, klasik ve moleküler tanı yöntemleri ile anastomosis grupları belirlenmiştir. Elde edilen 34 Rhizoctonia solani izolatının AG-1 IB (1 izolat), AG-2-1 (4 izolat), AG-4 (HGI, HGII ve HGIII alt gruplarına ait 24 izolat) ve AG-5 (5 izolat); iki çekirdekli Rhizoctonia izolatlarının ise AG-E (2 izolat) ve AG-K (2 izolat) olduğu saptanmıştır. Fasulye yapraklarında yapılan patojenisite testinde AG-1 IB’nin en virülent grup olduğu, AG-4 ve AG-5 izolatlarının sırasıyla onu takip ettiği belirlenmiştir. Baklada ise AG-1 IB ve AG-4’e ait izolatların virülensliğinin yüksek olduğu görülmüştür. Diğer anastomosis gruplarına ait izolatlar ise yaprakta veya baklada genellikle enfeksiyon oluşturmamıştır. Türkiye’de bugüne kadar tarla şartlarında fasulye bitkilerinin toprak üstü aksamlarında Rhizoctonia tür ve anastomosis gruplarının belirlenmesine yönelik yapılan ilk çalışmadır.

Anastomosis Groups and Pathogenicity of Rhizoctonia Species Isolated from Bean Pods in Erzincan Province

This study was carried out to determine the anastomosis groups (AG) and pathogenicity of Rhizoctonia isolates that cause web blight on the aerial parts of bean plants in Erzincan province. At the result of the study, totally, 38 Rhizoctonia isolates were obtained from bean pods, and their anastomosis groups were identified using classical and molecular techniques. Thirty-four Rhizoctonia solani isolates were identified as AG-1 IB (1 isolate), AG-2-1 (4 isolates), AG-4 (24 isolates belonging to HGI, HGII and HGIII subgroups) and AG-5 (5 isolates); binucleate Rhizoctonia isolates were identified as AG-E (2 isolates) and AG-K (2 isolates). In pathogenicity test on bean leaves, the most virulent group was determined as AG-1 IB, it was followed AG-4 and AG-5 isolates, respectively. On bean pods, the virulence of AG-1 IB and AG-4 isolates was found to be high. On the other hand, isolates belonging to other anastomosis groups generally did not cause infection on leaves or pods. In Turkey, the anastomosis groups of Rhizoctonia isolates obtained from the aerial parts of bean plants in fields were determined for the first time in this study.
