Buğday (Triticum durum Desf.) ve arpa (Hordeum vulgare L.) in vitro fidelerinin bor alımının ICP-AES ile tespiti

Bu araştırma, ICP-AES (Inductively Coupled Plasma-Atomic Emission Spectrometry) ile buğday (Triticum durum Desf.) ve arpa (Hordeum vulgare L.) in vitro fidelerinin bor (B) alımı ve biriktirme durumlarını belirlemek amacıyla yürütülmüştür. Tohumlar; 200 ml 'lik cam kavanozlarda, %0.7 ağar, %3 sakaroz ve sırasıyla; 0.0, 6.2, 18.6, 55.8, 111.6 mg/l $H_3BO_3$ içeren 50 ml MS besin ortamında, her kavanozda 5 adet tohum olacak şekilde kültüre alınmıştır. Yirmi günlük fidelerin kurutulmuş kök ve gövde kısımları (0.1-1 g) ile yetiştirme ortamları (5.0-6.0 g) mikrodalgada (CEM-Mars x 5) 10 mi $HNO_3$ ile 170 PSI basınçta 200°C'de 40 dak. yakılmış ve numunelerde ICP-AES ile bor analizi yapılmıştır. Kızıltan-91 'de köklerde en az B birikimi 2.2$\mu$g/g kuru madde olarak kontrol ortamından elde edilirken, en çok kök B içeriği ise 15.1 \mu$g B/g ile 111.6 mg/L $H_3BO_3$ içeren ortamdan elde edilmiştir. Gövdede B birikimi en az 4.9 $\mu$ g B/g ile kontrolde, en çok 67.6 $\mu$ g B/g ile 111.6 mg/l $H_3BO_3$ içeren ortamında olmuştur. Tolcak-157/37'de en düşük kök B konsantrasyonunun 0.53 $\mu$ B/g ile yine kontrol ortamında, en yüksek birikimi ise 17.3 $\mu$B/g ile 111.6 mg/l $H_3BO_3$ de olmuştur. Arpada en düşük gövde B konsantrasyonunun 0.74 $\mu$g B/g ile 0 mg/l $H_3BO_3$da, en yüksek 100.5 $\mu$g B/g ile 111.6 mg/l $H_3BO_3$ da yetiştirilen fidelerin gövdelerinde biriktiği belirlenmiştir.

Boron uptake of in vitro seedlings of wheat (Triticum durum desf.) and barley (hordeum vulgare L.) as determined by ICP-AES

Boron (B) absorption of in vitro seedlings of wheat (Triticum durumDesf.) and barley (Hordeum vulgareL.) and its accumulation in various organs were investigated using an Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES). Dried plant samples (0,1-1 g) from 20-d old seedlings and plant culture media (5.0-6.0 g) were transferred into plastic bottles containing 10 ml of 65 % (v/v) nitric acid $HNO_3$ solution. Samples were ashed at 200°Cfor 40 min in a microwave (CEM-Mars x 5) followed by analyses in the ICP-AES device. The lowest concentration of B (2.2 $\mu$g B/g dry matter) accumulation was found in roots of Kızıltan-91 seedlings cultured in the control medium containing 0 mg/l B -whereas the highest B accumulation (15.1 /$\mu$ g B/g dry matter) was detected again in roots of Kızıltan-91 in medium containing the highest concentration of B that was incorporated into the medium as $H_3BO_3$. Similarly, the highest level of 8 absorption by the shoots of Kızıltan-91 took place in media containing the highest level of boric acid (111.6 mg/l) with the lowest B concentration was found in shoots at the lowest medium B concentration (0 mg/l), showing that there was a linear correlation with respect to B content of the culture medium and B accumulation in the organs of Kızıltan-91. The B absorption response of barley seedlings were similar to that of wheat.
