Amaç: Yapılan in vitro çalışmalarda, metilen mavisinin (MM) iskemi/repeıfüzyon sonrası ortaya çıkan serbest oksijen radikallerini, ksantin oksidaz enzimi üzerine alternatif bir elektron yakalayıcı rolü oynayarak azalttığı gösterilmiştir. Bu çalışmada testis torsiyonu modelinde, detorsiyondan önce intraperitoneal verilen MM'nin reperfüzyon hasarı üzerine etkisi incelendi. Yöntem: Çalışmada 10 denekli dört grup SpragueDawley cinsi sıçan kullanıldı. Grup1 sham, diğer üçü tedavi grubu olarak düzenlendi. Bir saatlik torsiyon döneminde, detorsiyondan 10 dakika önce Grup 2'de serum fizyolojik (SF), Grup 3'de allopurinol (AL) ve Grup 4'te MM verildi. Aim: In vitro studies have demonstrated that methylene blue (mb) acts as an alternative electron acceptor for xanthine oxidase enzyme and suppresses the production of Superoxide radicals o2 in reperfusion injury. In this study, the effect of intraperitoneal mb on reperfusion injury was investigated in a testicular torsion model. Method: SpragueDawley rats were divided into four groups, each consisting 10 rats. In group I, sham operation was performed. In other three groups after 60 min torsion and W min before detorsion of the right testis, only isotonic saline (IS) in group II, Allopurinol (AL) in group III and MB in group IV were injected intraperitoneally. Following the 60 min detorsion period, blood sample were obtained and orchiectomy was performed. Serum uric acid (UA) and myeloperoxidase (MPO) level and testis histopathology were evaluated. Results: UA was significantly lower in AL group compared with the other groups p<O.OO1 For MPO, while the difference was significant between AL group and the others (p<0.05), no difference was determined between IS and MB groups. Histopathological findings were evaluated with $x^2$ test. No difference was determined between AL and MB and also Sham groups. But the difference was significant between AL and IS groups. In AL group, injury spectrum was toward the low grades. There was no significant difference between MB and the others, but when compared to group IS, grade III injury was encountered less in MB group. Conclusion: According to these results, it could be concluded that in experimental torsion of the testis, injection of MB before detorsion does not significantly decrease the reperfusion injury of the testis and the inflammatory response, hut it may reduce the severity of histopathological changes.
