Domates güvesinin [Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae)] Batı Akdeniz Bölgesi populasyonlarının mitokondrial cytochrome oxidase subunit I (mtCOI)’e göre genetik varyasyonunun incelenmesi

Bu çalışmada Tuta absoluta`nın genetik varyasyonu mtCOI gen bölgesinin dizi analizine göre belirlenmiştir. Bu amaçla zararlının ergin bireyleri Batı Akdeniz bölgesinden Anamur, Gazipaşa, Serik, Antalya-Konyaaltı, Kumluca, Demre, Kınık ve Fethiye popülasyonlarından denemelerde kullanılmak üzere 2011 yılında domates bitkisinden toplanmıştır. Tek bir bireyden DNA izolasyonları yapılmış ve her bir popülasyondan en az 10 bireyden DNA izolasyonu yapılmıştır. DNA`lar mtCOI gen bölgesine spesifik “C1-J-2195 Forward ve TL2-N-3014 Reverse” primerleriyle çoğaltılmıştır. Sekans analizi sonunda yaklaşık olarak 747 bazlık sekans dizilimi Mega 4 programı ile dizi doğrulaması yapılarak analiz edilmiştir. Analizler sonucunda popülasyonlar arasında ve popülasyon içindeki bireyler arasında genetik farklılığın olmadığı belirlenmiştir. Gen bankasından elde edilen verilere göre Türkiye popülasyonları ile Güney Amerika ve bazı Avrupa popülasyonları arasında yüksek benzerlik olduğu gözlemlenmiştir. Sekans verileri ayrıca gen banka kaydedilmiştir.

Determination of genetic variation of tomato leafminer [Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae)] populations from west mediterreanean region of Turkey based on mitochondrial cytochrome oxidase I (mtCOI)

In this study, the genetic variation of Tuta absoluta was studied based on the sequences of mtCOI gene region. For this purpose, the samples of the adult pest were collected from tomato plants grown in Anamur, Gazipaşa, Serik, Antalya-Konyaaltı, Kumluca, Demre, Kınık and Fethiye district of western Mediterranean region in 2011. Minimum ten DNA was extracted individually from each. The mtCOI gene was amplified and sequenced with the primer pair “C1-J-2195 and TL2-N-3014 specific to this region. The sequences were aligned and analyzed by using Mega 4 software program. An approximately 747 base pair of informative sequences were obtained and used in the analyses. The results of the analysis showed that there was no inter and intra-population polymorphism in the sequences of individuals among the populations studied here. A high sequence similarity was found when the sequences of Turkish populations were compared with the Gen bank sequences of the South American and some European populations of the pest. Sequences were deposited in Gene bank.
