Karaciğer Kanser Hücrelerinin Sitoplazmik Özellikleri ile Kültür Süreci İlişkisi: Konfokal Mikroskopik Karşılaştırmalı Kantitatif Bir Çalışma

Hepatosellüler karsinoma tanısı konulmuş bir hastadan izole edilmiş olan HepG2 hücrelerinin morfolojik özelliklerindeki sitoplazmik organellerden mitokondri, endoplazmik retikulum ve golgi cisimciği ile hücre iskeletini oluşturan başlıca filament tipi olan aktin filamentlerinin miktarının değişimin kültür süreci ile bağlantılı olarak değişiminin karşılaştırmalı olarak değerlendirilmesi amaçlanmıştır. Gereç ve Yöntem: 37 C'da, %5 CO2 içeren inkübatörlerde, %10 Fetal Sığır Serumu FBS ile %1 Streptomisin-Penisilin içeren DMEM Dulbecco's Modified Eagle Medium içerisinde HepG2 hücrelerinin kültürleri yapıldı. 10 gün boyunca gözlenen bu hücreler; kültür sürecinin üç ayrı döneminde kültür yüzeyinin %30, %60 ve %100'ünü kapladıkları dönemlerde fikse edildi. İndirekt immunofloresan boyama tekniği ile ilgili sitoplazmik yapılar işaretlendi. İşaretli alanlar Zen ve Image J görüntü analiz programları kullanılarak ölçüldü. Sayısal sonuçlar grafiksel olarak ve Excel veri çözümleme aracı kullanılarak karşılaştırıldı. Sonuçlar regresyon ve eşleştirilmiş iki grup arasında t-testi farklı varyanslar varsayarak yöntemi kullanılarak karşılaştırıldı. Bulgular: Kanser hücrelerinin mitokondri, golgi cisimciği ve endoplazmik retikulum içeriğinde anlamlı bir değişiklik gözlenmesine karşılık; sitoplazmik aktin filament varlığımda belirgin bir farklılık gözlenmemiştir. Sonuçlar: Karaciğer kanser hücrelerinin sitoplazmik organel içeriği klasik kültür ortamında kültür sürecine, ortamdaki hücre yoğunluğuna bağlı değişmektedir. Hücre iskeletinde belirgin bir değişiklik olmamaktadır. Deneysel çalışmalarda; “çoğalma” aşamasındaki kültür hücrelerinin morfolojik özelliklerinin bu parametreler karşısındaki değişimi deneyelerin sonuçlarını etkileyebilir. Bu nedenle, araştırmalarda hücrelerin çoğalma aşamasınn hangi devresinde oldukları göz ardı edilmemelidir.

The Relationship Between the Hepatocellular Carcinoma Cells and the Cell Culture Duration: A Quantitative Comparative Confocal Misroscopic Study

It was aimed to compare the changes of morphologic features the content of mitochondria, endoplasmic reticulum, golgi apparatus and the actin filaments of the cell of the HepG2 cells, a cell line isolated from a patient having hepatocellular carcinoma, with the cultivation period. Materials And Methods: HepG2 cells were cultured in incubators 37 C, 5% CO2 within DMEM Dulbecco's Modified Eagle Medium including 10% Fetal Bovine Serum FBS and 1% Streptomycine-Peniciline,. The cells had been observed for 10 days and fixed when they covered 30%, 60% of 100% of the culture surface. Indirect immunoflourescence staining method had been used to label sitoplasmic contents. The labeled areas were measured by Zen and Image J image analysis system. The measurements were graphically visualized.The Excel data analysis tool wass used for the statistical analysis. The results were compared by using the regression method and t test. Results: Mitochondria, endoplasmic reticulum and golgi content of the cells changed significantly. However amount of the cytoplasmic actin filament in the cells remained similar. Conclusions: Cytoplasmic organels of the liver cancer cells changes according to the in vitro culture duration and the cell density. No dramatic change is seen at the structure of the cell cytoskeleton. Those changes of the cells at “proliferation” stages might effect the experiments. The current study demonstrates that the stage of the cultured cells during proliferation might be crucial to determine the results of the the studies.
