Kiraz Tomurcuklarında Kültür Başlatılmasına Bazı Sitokininlerin Etkisi

Bu çalışmada, kirazın lateral tomurcukları kullanılarak kültür başlatılmasına, sitokininlerden benzilaminopurin (BAP), thidiazuron (TDZ) ve kinetinin bazı konsantrasyonlarının etkisi üzerinde çalışılmıştır. Tomurcuklar, 0900-Ziraat çeşidine ait 5 yaşındaki ağaçların bir yaşlı dallarından dormant dönemde alınmıştır. Araştırmada BAP ve kinetinin 0.1, 0.5, 1.0, 2.0 ve 4.0 mgl-1 konsantrasyonları incelenmiş, TDZ’de bunlarla beraber 0.05 mgl-1 konsantrasyonuna yer verilmiştir. Kiraz tomurcuklarından kültür başlatmak için kinetinin 4 mgl-1 konsantrasyona kadar besi ortamına ilavesinin olumlu bir etkisinin olmadığı anlaşılmıştır. Kullanılan sitokinin tip ve konsantrasyonları içinde en başarılı sonuçlar 2 mgl-1 BAP ve 2 mgl-1 TDZ ilaveli besi ortamlarından alınmıştır. Ancak, TDZ bulunan besi ortamlarında gelişen rozet bitkilerin canlılığı, kültür süresinin sonuna doğru azalmıştır. BAP içeren besi ortamlarındaki kültürlerin daha sağlıklı ve canlı olduğu tespit edilmiştir. Besi ortamında 2 mgl-1 BAP içeren kültürlerde, %80.0 ± 9.2 oranında eksplantın alt kültüre alınabildiği, %43.8 ± 12.8 oranında rozet sürgün elde edildiği tespit edilmiş ve ortalama yaprak sayısı 1.71 ± 0.36 adet olarak belirlenmiştir.

Effect of Some Cytokinins on the Culture Initiation of Cherry Buds

In this study, the effect of different concentrations benzylaminopurine (BAP), thidiazuron (TDZ) and kinetin on culture initiation from cherry lateral buds was investigated. Buds was taken from one-year branches during the dormant period of 5-year-old trees belonging to 0900-Ziraat variety. Concentrations of 0.1, 0.5, 1.0, 2.0 and 4.0 mgl-1 of each of the three cytokinins were used and in addition to these concentrations, 0.05 mgl-1 was used for TDZ. It has been understood that the addition of kinetin to culture medium up to a concentration of 4 mgl-1 does not have a positive effect in order to initiate culture from cherry buds. The most successful results in the types and concentrations of cytokinins used were taken from 2 mgl-1 BAP and 2 mgl-1 TDZ supplemented media. However, the viability of rosette plants grown in TDZ containing media has decreased towards the end of the culture period. In the BAP supplemented medium, more vigorous and healthy rosette shoots were obtained. In cultures containing 2 mgl-1 BAP in the medium, the rate of subculture explants was 80.0 ± 9.2%, the rosette shoot rate was 43.8 ± 12.8% and the number of leaves was 1.71 ± 0.36.
