Çeşitli Gıda Boyalarının Hücre Canlılığı Üzerine Etkisinin MTS Testi Kullanılarak Belirlenmesi

Renk, gıdanın kolayca tanınması ve nihai kararın verilmesi için günümüz modern toplumunda vazgeçilmez bir özelliktir. Gıda boyalarının toksikolojisi, özellikle boyar maddelerin kansere sebep olabileceği konusu gündeme geldiğinden beri birçok araştırma yapılmış ve gıda boyalarının önemli kısmı ulusal ve uluslararası onaylanmış listelerden mutajenik ve kanserojenik aktivitelerinden dolayı çıkarılmıştır. Bu çalışmada, esası hücre canlılığını belirlemeye dayalı olan MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium assay)  testi kullanılarak Tartrazin, Eritrosin, New Coccine, Rose Bengal, Pholoxine, Allura Red ve Amaranth sentetik gıda boyalarının toksiditelerinin ölçülmesi amaçlanmıştır. Adı geçen her bir boyanın 1mM konsantrasyonları ile CHO (Chinese hamster ovary) hücreleri inkübe edilmiştir. MTS testinin sonucuna göre Eritrosin, Rose Bengal, Pholoxine, Allura Red ve Amaranth boyalarının kontrole göre hürce canlılığı üzerine istatistik olarak önemli derecede (p<0.05) toksik olduğu tespit edilmiştir.

Determination of Effect of Various Food Colorants on Cell Liveliness Using MTS Test (Turkish with English Abstract)

Color, an inevitable feature used to easily recognize and also to reach final decision about the food in our modern world. Since issue about toxicology of food color agents especially their carcinogen properties has received interest, several research studies have been done and according to their results important parts of food colors were taken out from the food additives lists approved by national and international agents. In the present study, MTS assay (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium assay)  determining of vitality of cells was used to measure toxicity of several artificial food colors (Tartrazine, Erythrosine, New Coccine, Rose Bengal, Pholoxine, Allura Red and Amaranth). One mM concentration of each color was incubated with CHO (Chinese hamster ovary cell). According to MTS assay,  toxicity of Erythrosine, Rose Bengal, Pholoxine, Allura Red and Amaranth against cell vitality was evaluated significantly higher comparing the control (p<0.05)
