Biyolüminesans yöntemi ile herhangi bir örnekteki mikroorganizma sayısının tahminlenmesi ateş böceklerinde görülen lusiferin-lusiferaz reaksiyonuna bağlı hızlı ve duyarlı bir tekniktir. ATP biyolüminesans yönteminde en fazla 1 saat içinde sonuç alınabilmektedir. Ancak bu yöntemde bazen gıda orijinli somatik ATP, mikrobiyal ATP’nin saptanmasında bazı interferanslara neden olmakta ve sağlıklı sonuçlar elde edilememektedir. Bu yöntemle spesifik bir takım mikroorganizmaların ve patojenlerin analizinin de yapılabilmesi için şu anda mevcut çalışmaların dışında değişik çalışmalar yapılmalıdır. 

Bioluminescent measurement of microbial ATP using the firefly luciferin-luciferase system is a rapid and sensitive technique for the estimation of microbial populations which may be completed within 1 hr. This represents considerable time-savings in comparison to conventional microbial enumeration techniques requiring 2-3 days for completion. It is reported that there are some difficulties in applying microbial ATP measurements to foods including interference from ATP of nonmicrobial origin. Various developments are necessary to increase the acceptance of the method, in particular automation and the provision of more detailed information such as analysing of some pathogenic and specific microorganisms.
