Tam büyüklükteki hepatitis B virüs genomunun polimeraz zincir reaksiyonuyla gösterilmesi

Amaç: Hepatitis B virüsü (HBV) hakkındaki çalışmalar polimeraz zincir reaksiyonuyla (PZR) çoğaltılan subgenomik ya da tam uzunluktaki HBV genomu kullanılarak yapılmaktadır. Hepatitis B virüsü hücre kültürferinde üretilememektedir. Tam uzunluktaki HBV genomu klonlama vektörlerine yerleştirilerek düşük litrede olsa bile-hücre kültürlerinde üretilebilmekte ve aynı zamanda HBV genomu üzerinde çalışmalar yapılabilmektedir. Yöntem: Bu çalışmada 15 kronik hepatitis B hastası çalışma kapsamına alınmış ve 9 hastanın 3200 baz çifti uzunluğundaki HBV genomunun PZR ile çoğaltılması optimize edilmiştir. Bulgular ve sonuç: Enzim kesim deneyi yapılarak çoğaltılan PZR ürünlerinin HBV genomuna ait olduğu tespit edilmiştir.

Detection of whole hepatitis B virus genom with polymerase chain reaction

Objective: Current knowledge of hepatitis B virüs (HBV) is based on either amplified subgenomic HBV fragments or whole HBV genomes. HBV can not be proliferate in cell culture. However, whole HBV genome placed into cloning vector proliferates in the celi culture at low level titreş and allows us to research on the HBV genome. Methods: Fifteenth chronic hepatitis B patients included in this study. Whole HBV genome 3200 base pair in length was amplified by polymerase chain reaction (PCR) in 9 patients. Results and conclusion: Enzyme digestion assay indicated that the PCR products belong to HBV genome.
