Dactylorhiza osmanica’nın Topraküstü Kısımlarında Antioksidan, Antimikrobiyal ve Tirozinaz İnhibitör Aktivitelerinin Araştırılması

Amaç: Ülkemiz zengin florasıyla çok sayıda tıbbi ve aromatik bitkiyi bünyesinde barındırmaktadır. Günümüzde fitoterapi araştırmaları popüler olmuştur. Bu çalışmanın amacı, Dactylorhiza osmanica bitkisinden elde edilen metanolik ekstrenin antioksidan ve antimikrobiyal kapasiteleri, tirozi-naz inhibitor etkisini değerlendirmektir. Gereç ve Yöntem: Ekstrenin antioksidan özelliği toplam fenolik içeriği (TPC), demir indirgeyici antioksidan güç (FRAP) ve 1,1-difenil-2-pikrilhidrazil (DPPH) radikali süpürme aktivitesi testi gibi in vitro antioksidan tayin yöntemleri kullanılarak araştırıldı. Ekstrenin fenolik kompozis-yonu ters faz yüksek performanslı sıvı kromatografisi (RP-HPLC) ile incelendi. Antimikrobiyal aktivite disk difüzyon metodu kullanılarak test edildi. Tirozinaz inhibitör aktivite kolorimetrik olarak ölçüldü. Bulgular: Ekstrenin TPC, FRAP ve DPPH aktiviteleri sırasıyla; 20.6 ± 0.3785 mg gallik asid eşdeğeri/gram numune, 804 ± 8.6217 μM Trolox/g numune ve 0.1838 ± 0.0015 mg/mL olarak bulundu. Gallik asit, protokatekuik aldehit, protokatekuik asid, p-hidroksi benzoik asit, klorojenik asit, vanilik asit, kafeik asit, vanilin, şiringaldehit, p-kumarik asit, ferulik asid, sinapik asid, benzoik asid ekstrede bulunan fenolik bileşiklerdir. Tirozi-naz inhibitör aktivite tayininde ekstrenin IC50 değeri anlamlı bulunmamıştır. Ekstre, asid-hızlı bakteri (M. smegmatis), bazı gram pozitif (S. aureus and B. cereus) ve bazı gram negatif (Y. pseudotuberculosis) bakterilere karşı orta derecede antibakteriyal aktivite gösterdi. Sonuç: Bu çalışmanın sonuçlarına göre, D. osmanica yeni farmasotiklerin geliştirilmesinde potansiyel bir kaynak olarak düşünülebilir.

Investigation of Antioxidant, Antimicrobial and Tyrosinase Inhibitor Activities on the Aerial Parts of Dactylorhiza osmanica

Objective: Our country contains a large number of medical and aromatic plants with rich flora. Nowadays phytotherapy research has become popular. The purpose of this study was to evaluate the antioxidant and antimicrobial capacities, and tyrosinase inhibitory effect of methanolic extracts of Dactylorhiza osmanica. Material and Method: The antioxidant properties of extract were investigated using in vitro antioxidant assay methods such as total phenolic content (TPC), ferric reducing antioxidant power (FRAP), and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity assay. Enzyme inhibi-tory effect was tested aganist tyrosinase. The phenolic composition of extract was investigated by means of reverse phase-high performance liquid chromatography (RP-HPLC). Antimicrobial activity was tested using the disc diffusion method. Tyrosinase inhibitor activity was tested to colorimet-rically. Results: The total phenolic content value is 11.2 ± 0.472 mg gallic acid per gram sample, FRAP value is 240 ± 2.645 μM Trolox/g sample, and IC50 value for DPPH assay has been found as 0.708 ± 0.010 mg/mL. Phenolic compounds were identified as gallic acid, proto-catechuic acid, proto-catechuic aldehyde, p-hydroxy benzoic acid, chlorogenic acid, caffeic acid, vanillin, p-coumaric acid, syringaldehyde, ferulic acid, and benzoic acid. The IC50 value of the extract was not significant in determining tyrosinase inhibitor activity. The extract displayed moderate antibacterial activity against an acid-fast bacterium (M. smegmatis), some Gram pozitive (S. aureus and B. cereus) and Gram negatif (Y. pseudotuberculosis) bacteria. Conclusion: According to the results of the present study, D. osmanica can be considered as a potential source for developing new pharmaceuticals.
