Köpeklerin Gençlik Hastalıği Aşısı ve Muafiyet Deneyleri

Köpeklerin gençlik hastalığı için Lederle avianze Distempeıvirusu ile bir aşı istihsal edildi.Bu maksat için 8 günlük embriyolu tavuk yumurtaları kullanıldı.Bunların chorioallantoic membranlarına 52. pasaj virusunun1/10 suıspanısiyonundan 0.2 ml inoküle edildi. Yurnurtalar 7 gün müddetle 37° C de inkubasyondan sonra açılarak membranları toplandı, ezildi, santrüfüj edildi. Üstteki kısım alınaraık % 20 nisbetinde her ml de 100 ünite penicilin ihtiva eden dayanıklılık verici ( % 2 pepton, % 10 laktoz, distile su PH 7.2) vasat ile sulandırıldı. Sterilite kontrolu ve yumurtada titrasyonu yapıldı. 2 ml miktarında şişeleretaksim edilerek liyofilize edildi.Virusun titri ID50 103•8 -  IDso 104 / 0.1 ml olark tesbit edildi.Bu çalışmada 2 aylıktan büyük 24 adet köpek yavrusu kullanıldı.Bunlardan 15 ine subkutan yol ile 2 ml aşı verildi. 6 sı eprüvasyonda kontrol olarak kullanıldı ve 3 adedi de deney süresince aşılı köpeklerle temasa bırakıldı.Aşılı köpeklerin 10 gün müddetle dereceleri alındı, herhangibir termik reaksiyon tesbit edilemedi.Köpeklerin hepsinden aşılamadan evvel ve aşılandıktan 1 ay, 6 ay ve 12 ay sonra serumları alınarak serum, nötralizasyon testine tabi tutuldu. Aşılı köpeklerden 5 adedi 3 kontrol ile heraber aşılamadan1 ay sonra Snyder Hill virusunun 1/10 dalak süsponsiyonundan 1 ml verilerek intravenöz yol ile eprüve edildi. 15 gün müddetle sabah - akşam dereceleri alındı. Aşılılarda bariz bir dereoe yükselmesi tesbit edilememekle beraber kontrollarda yüksek fievr ve hastalığın diğer belirtileri görüldü.Geri kalan 10 aşılı köpek 3 kontrol ile beraber vaksinasyondan 12 ay sonra yukarda anlatıldığı tarzda eprüve edildi. Üçüncü gün 2 aşılı köpekle beraber 3 kontrolda derece yükseldi ise de aşılılarda1 - 2 gün içinde normale düştü. Knntrollar:da hastalık belirtileri de başladı ve biri 10. gün Distemper'den öldü. Kontakt tecrübesinde bulunan 3 köpekte tecrübelerin devamı müddetince herhangi bir değişiklik tesbit edilemedi. ·Aşılı köpeklerin serumlarında 12. aya doğru antikor titresinin biraz düştüğü müşahede edildi.

THE PRODUCTION OF AN AVIANIZED CANINE DISTEMPER VACCINE ANP THE IMMUNITY TETSJN DOGS.

A vaccine was produceıd w.ith Lederle Aviaınized OD virus in order to immunize the dogs agecins-t Distemper.For the production of :the vaccine eggs that had ibeen embryonaıted for 8 days were each iocıoculated on the chori'oaıll.antoic membraırıewith 0.2 mıi of a: 10 percent suspeı:nsion of ı52nd paJS·sage mateııials of the egg - adapted OD virus.After an ineubatian period of 7 days at 37° C. chorioallantoicmembranes were collected and homogenized in a ahıilled blender-:These marterials were then centrifuged. The supernataırııt waıs co1~lected and was made into a 20 percent suspension with a medium(% 2 peptone, % ı O Lactose, dist. water PıH 7.2) cootaining ı 00uınit·s of pencil'lik.i p~r ml ..The titration ·İn e:ggs and the s-teriHty teısts were conducted.lt was then diJStributed in 2ml quanrtities in small vi·a:1s aınd lyophi~lized.The titers of viruıs in embryonated chicken eggs were IDso ıo3.8to I•Dso 10'' per O.ı ml.In the se studies 24 puppies aıbout 8. to 9. weeks _o.f . age w ereused. A group of ıs puppies were inoculated suıbcutarie,<>usly with 2ml of vacoine. Six conıtrols were used; and tıh.e other 3 pupp~eswere left as the contact controls along witıh tıhese studıies.Afte:r vaccination the temperatures were· tak-en daily from thevaccinated puppies for 10 days. None of these va:ccinated puppiesshowed any increaısed temperature.Serum - neutralization tesıts wereı carried out with the prevaccin:aıtion and postvaccıim:ation sera taıken at the intervals of ı,6 and12 months from all puppiesı.The virulent virus., Snyder Hill strain wa;s uısed for chaJ,lengeanıd 4 week-s af-ter vaccination 5 of the:se vacoinated dogs were challe-nıged intravenouıs1y with ı ml of a ıo percent suıspension of infectedsple:en. 3 cOilJtrols were uısed.Temperatures were taıken daily. for 15 days after the challengedogs. None of .the vaccinated .dogs· showed any 1ncreaıSed temperaturıe.AH ccintroilıs showed Mgh tempemt:ure ~ and the ·other .si:gns oıfillness.12 months after vaccin.ation the remaninıg 10 vaccinated dogsweıre challenged as the same manner deseribed aıbove using 3 controls.On the 3ııd day of the chaHenıgs two vacdna.ted ıdogs and aHcontrols showed high •temperature. Within two days the vaccinat·eddogs becaıme normal again. The controls. showed the signıs of CD.One control doıg died on the lOth day.A:long witlı these tests all vacdnated dogs; except 2, showed nosiıgns of illness and unvaccina:ted dogs pbıced in contact alsa remainedfree from OD.The positive logs of ·serum - tiıters determined on 1 O vaccinateddogs showed a slight decrease at the termination of tlıe · test.
