Kanatlı Kökenli Escherichia coli’lerin Filogruplandırılması
Filogruplandırma, Escherichia coli suşlarının karakteristik özelliklerinin anlaşılması, enfeksiyonların önlenmesi, kontrol edilmesi ve yeni tedavi yöntemlerinin belirlenmesinde önemli rol oynamaktadır. Bu çalışmada, kanatlı kökenli avian patojenik E. coli (APEC) ve avian fekal E. coli (AFEC) suşlarının filogruplandırılması amaçlandı. Çalışmada 150 adet APEC ve 150 adet AFEC suşunun filogruplandırılması kuadrupleks PCR metodu ile gerçekleştirildi. Çalışma sonucu 150 APEC suşunun 4’ü A (%2.66), 43’ü B1 (%28.66), 38’i B2 (%25.33), 2’si C (%1.33), 18’i E (%12), 5’i F (%3.33), 1’i Clade I (%0.66), 2’si negatif (%1.33) olarak belirlenirken 37 (%24.66) suş gruplandırılamadı. İncelenen 150 AFEC suşunun ise 5’i A (%3.33), 8’i B1 (%5.33), 16’sı B2 (%10.66), 16’sı C (%10.66), 1’i D (%0.66), 27’si E (%18), 9’u F (%6), 3’ü Clade I veya Clade II (%2) olarak gruplandırılırken 51 (%34) gruplandırılmadı. Escherichia coli enfeksiyonlarının önlenmesi amacıyla geliştirilecek olan aşılarda yersel suşların kullanılması ve bu suşlara ait özelliklerin belirlenmesi gerektiği bilinmektedir. Bu nedenle, elde edilen sonuçlar epidemiyolojik açıdan önem taşımaktadır. Bununla birlikte çalışma sonunda, genotipik olarak çeşitlilik gösteren E. coli suşlarından, özellikle APEC suşlarının, daha detaylı genotipik analizler yapılarak yeni filogrupların araştırılması gerektiği vurgulandı.
