Plazma glutamin düzeylerinin iki yöntem ile değerlendirilmesi

Kan plazması glutamin analizinde kullanılan ince tabaka kromatografisi ile kombine bir spektrofotometrik yöntem (İTK-SPK) bilinmektedir. Amacımız, bu yöntem ile ince tabaka kromatografisinde özel okuyucu (İTK-Okuyucu) yardımıyla geliştirdiğimiz yöntemi kıyaslamaktır. Bu amaçla, 35 sağlıklı bireyden kan alınıp plazmaları ayrılmıştır. Ekstraksiyondan sonra, ince tabaka kromatografisi ile ayrılmış glutaminin spektrofotometrik ve 'İTK-okuyucu' aletinde miktar tayinleri yapılmıştır. Sağlıklı kişilerden alınan kan plazma örneklerinin İTK-okuyucu ve spektrofotometrik yöntemle bulunan glutamin düzeyleri arasında uygunluk bulunmuştur (korelasyon katsayısı, r: 0.883).

The evaluation of plasma glutamine levels by two methods

It has been known a method used analysis of glutamine in blood plasma combined with thin layer chromatography (TLC-SPC). Our aim was to compare this method (TLC-SPC) with 'TLC-Scanner' procedure combined with thin layer chromatography developed by us. With this aim, we collected blood from 35 healthy volunteer controls and separated their plasma. After the extraction procedure, glutamine was separated by thin layer chromatography and quantative analysis was carried out in "TLC-Scanner" and spectrophotometer. Glutamine results, obtained by spectrophotometric procedure in plasma samples taken from healthy controls were complied with the results of TLC-Scanner' (correlation coefficient, r: 0.883).
