Parkinson hastalarında apolipoprotein E gen polimorfizminin lipid profilleri ile ilişkisi
Demans gruplarında yapılan pek çok çalışmada apolipoprotein E ε4 allelinin Alzheimer hastalığının erken ve geç başlangıçlı formu ile ilişkili olduğu ileri sürülmektedir. Apolipoprotein E'nin lipid metabolizmasında çok önemli görevleri vardır. Yapılan çalışmalarda homozigot apolipoprotein E ε3 alleli taşıyan bireylerle karşılaştırıldığında ε2 allelini taşıyan bireylerin serum total kolesterol (TC) ve düşük dansiteli lipoprotein kolesterol (LDL) seviyelerinin daha düşük olduğu; ε4 alleli taşıyan bireylerde ise daha yüksek olduğu gösterilmiştir. Bu bulgulara rağmen farklı toplumlarda apo E allellerinin serum lipid konsantrasyonları üzerine etkileri hala netlik kazanmamıştır. Çalışmamızda 21 Parkinson hastasında ve 29 sağlıklı yaşlı bireyde apolipoprotein E gen polimorfizmi ile birlikte serum lipid profilini inceleyerek bu hasta grubunda apolipoprotein E gen polimorfizmine bağlı olarak gelişen lipid profili değişimlerini incelemeyi amaçladık. Çalışmada apo E genotipini belirlemek için polimeraz zincirleme reaksiyonu (PCR) ve agaroz jel elektroforez teknikleri kullanıldı. Serum lipid profilleri enzimatik yöntemlerle tayin edildi. Apo ε4 allel frekansı hasta grubunda 0.047 iken kontrol grubunda 0.017 olarak tespit edildi. The association between apolipoprotein E gene polymorphism and lipid profiles in patients with Parkinson's disease
Apolipoprotein ε4 has been associated with both early and late-onset Alzheimer's disease (AD). Several studies of the effects of apoE polymorphism on lipid metabolism have shown that individuals with the ε2 allele tend to have lower levels of serum total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) than do individuals who are homozygous for the ε3 allele, whereas individuals with the ε4 allele tend to have higher levels of serum TC and LDL-C than do ε3 homozygous . Despite this consensus, questions remain as to whether the biological effects of different apoE alleles on serum lipid concentrations are the same across different populations. Because dementia in AD and Parkinson's disease (PD) share many biologic and clinical features, we determined the apo E genotypes and lipid profiles for 21 patients with PD and 29 age-matched healthy elderly controls from same community. PCR (Polymerase Chain Reaction), RFLP (Restriction Fragment Length Polymorphism), and agarose gel electrophoresis techniques were used to determine the apo E genotypes.
