Kişniş (Coriandrum sativum L.) uçucu yağ ekstresinin letal doz düzeyleri ile antienflamatuvar aktivitesinin araştırılması

Bu çalışmada, kişniş (Coriandrum sativum L) (CS) uçucu yağ ekstresinin farelerde letal doz düzeyleri ile sıçanlarda carrageenan'la oluşturulmuş sağ-arka pençe ödemi modelinde antienflamatuvar etkinliğinin araştırılması amaçlanmıştır. Serum fizyolojik kontrol, etil alkol kontrol, indometazin (3 mg/kg) referans grupları ile CS (0.010 ml_/kg) ve CS (0.020 mL/kg) grupları oluşturuldu. Sıçanların sağ arka-pençeleri pletismometre aracılığı ile ilaç uygulamasından önce ölçüldü. Ardından ilaç periton içi yolla verildi. Subplantar yolla lambda-carrageenan uygulandı ve bu uygulamadan üç saat sonra sağ-arka pençe hacmi tekrar ölçüldü. Her iki ölçüm arasındaki fark gruplar arasında istatistiksel olarak karşılaştırıldı. Çalışma sonunda indometazin'in % 95.70, CS (0.010 mL/kg) ekstresinin % 46.97 ve CS (0.020 mL/kg) ekstresinin % 53.16 oranında antienflamatuvar etkinlik gösterdiği saptandı. İstatistiksel olarak her iki doz CS ekstresinin kontrol grubuna göre anlamlı seviyede antienflamatuvar etkinlik gösterdiği, bu etkinin indometazin'den daha zayıf olduğu tespit edildi. CS uçucu yağ ekstresinin LD50 seviyesi 2.257 ml_/kg olarak hesaplandı. Sonuç olarak, CS uçucu yağ ekstresinin indometazinden daha zayıf bir antienflamatuvar aktiviteye sahip olduğu kanaatine varıldı.

The levels of lethal dose and anti-inflammatory effect of Coriandrum sativum L.essential oil extract

The levels of lethal dose of Coriandrum sativum L. (CS) essential oil was studied in mice and anti-inflammatory effect of CS essential oil was studied with carregeenan-induced hind-paw edeme model in rats. The study involved 5 groups; Serum physiologic, ethyl alcohol, indomethacin (3 mg/kg), CS (0.010 mL/kg), CS (0.020 mL/kg). After measuring the volumes of right hind-paws of rats using a plethysmometer, drugs were injected intrapehtoneally and lambda-carrageenan were injected subcutaneously into the plantar region. Three hours after the injections the volume measurements of the right hind-paws were repeated and the difference between the volumes were compared. It was found that indomethacin reduced the inflammation by 95.70 %, 0.05 mUkg CS by 46.97 % and 0.10 mUkg CS by 53.16. %. Both doses of the CS exctract used showed statistically significant anti-inflammatory effect compared to the control groups. The anti-inflammatory effect of both doses of CS was weaker than the effect of indomethacin. The median lethal dose (LD5o) of the extract of CS was determinated as 2.257 ml/kg. As a result CS essential oil extract was shown to have an anti-inflammatory effect.
