Amniyotik sıvı hücre kültüründe açık ve kapalı sistemlerin değerlendirilmesi

Bu çalışmada hücre kültürlerinin açık ve kapalı sistemlerdeki üremeleri değişik besiyerleri kullanılarak karşılaştırıldı. Hücre kültürleri kullanılan tüp ya da flask başına maliyet, çalışma zamanı, mitotik indeks açılarından incelendi. Sırasıyla gelen 100 amniyotik sıvı örneğinden hem açık, hem de kapalı sistem kullanılarak hücre kültürü yapıldı. Açık sistemde hücre kültürleri için 25 cm2'lik doku kültür flaskı, %5 CO2 ve %95 nemlendirilmiş hava karışımını sağlayacak inkübatörler, kapalı sistemde, 10 mL lik altı düz tüpler ve kuru etüvler kullanıldı. Açık sistemde penisilin/streptomisin ve L-Glutamin eklenmiş BioAmf, kapalı sistemde Hepes, L-Glutamin, penisilin-streptomisin, ultroser-G ve fetal calf serum eklenmiş F-10 "Ham's Nutrient Mixture" ile "Chang-D" karışımı besiyeri kullanıldı. 100 amniyotik sıvı örneğinden kapalı sistemde kültürü yapılanların tamamında hücre üremesi saptandı. Bunlardan bir tanesinde hücre üremesi saptanmasına rağmen yeterli sayıda ve kalitede metafaz elde edilemedi. Açık sistemde kültür yapılanların dördünde mantar enfeksiyonu gelişti. Yirmisekiz örnek çeşitli nedenlerle çalışmadan çıkarıldı. Çalışmanın yapıldığı dönem içinde açık sistemde bir flask başına yapılan harcama, kapalı sistemde bir tüp çalışmaya hazır hale gelinceye kadar tüp başına yapılan harcamaya göre 4 misli daha pahalı olarak bulundu. Açık sistemde ortalama mitotik indeks 10.46±12.76; kapalı sistemde 9.16±9.75 bulundu (p>0.05).
