Samsun ilinde Beet necrotic yellow vein virus ve/veya Beet soilborne virus ile bulaşık şeker pancarı üretim alanlarında virüs taşıyan Polymyxa betae Keskin oranının belirlenmesi

Bu çalışma, Samsun ili şeker pancarı (Beta vulgaris L.) üretim alanlarına ait toprak örneklerinde ‘En Yüksek Olası Sayı' (Most probable number: MPN) tekniği ile Beet necrotic yellow vein virus (BNYVV) ve/veya Beet soilborne virus (BSBV) ile bu virüslerin vektörü olan Polymyxa betae'nın inokulum yoğunluklarının belirlenmesi ve viruliferous (virüs içeren) P. betae oranlarının ortaya konulması amacıyla yürütülmüştür. Bu amaçla, BNYVV'ye hassas şeker pancarı çeşidi bölgeden seçilen topraklara ekilmiş ve sonrasında virüslerin tespiti için ELISA ve P. betae için de asit fuksin ile kök boyamasını takiben ışık mikroskobu kullanılarak analizler yapılmıştır. Araştırma sonucunda; BNYVV ile bulaşık Bafra ve Vezirköprü toprak örneklerinde viruliferous P. betae oranları %0.4 ile %1.2 olarak belirlenirken, BSBV ile bulaşık Havza ve Kavak ilçelerinde ise bu oranın %9.7 ile %17.3 olduğu tespit edilmiştir. Bununla birlikte, her iki virüs ile bulaşık olan Vezirköprü ilçesi örneğinde virüs içeren P. betae'ların %4.8'inin BNYVV, %5.7'sinin ise BSBV ile bulaşık olduğu saptanırken; Bafra toprak örneğinde bu oranın BNYVV için %54.7, BSBV için %66.7 olduğu belirlenmiştir.

Determination of proportion of viruliferous Polymyxa betae Keskin in sugar beet production areas infested with Beet necrotic yellow vein virus and/or Beet soilborne virus in Samsun province

This study was conducted to determine P. betae, BNYVV and/or BSBV inoculum densities in sugar beet (Beta vulgaris L.) production areas infested with soilborne viruses in Samsun province by Most probable numbers (MPN) technique and to determine the percentages of viruliferous P. betae. For this purpose, susceptible sugar beet cultivar to BNYVV was grown in the collected soil samples and then analysed using Enzyme-linked immunosorbent assay (ELISA) for virus detection, and using light microscopy following roots staining with acid fuchsin for detection of P. betae. The result of this study showed that 0.4% and 1.2% of the infective population of P. betae were viruliferous in BNYVV infested soil samples from the Bafra and Vezirkopru districts, respectively. Also, in BSBV infested soils from Havza and Kavak districts, 9.7% and 17.3% of the infective population of P. betae were viruliferous. In mixed infested soil samples from Vezirköprü, the percentages of viruliferous P. betae were 4.8% for BNYVV and 5.7% for BSBV. In another soil with mixed virus infection from Bafra, 54.7% and 66.7% of the infective populations of P. betae were viruliferous for BNYVV and BSBV, respectively.
