Dllhasf)nra belirli aralıklarla alınan selefon diskler, üzerlerindeki fungus ile blrlikte periodikasit Schiff yöntemi veya analin mavisi ile boyanıp, eti! alkol serisi vexylol dengeçirildikten :Sonra Kanada balzamı, Euparal veya Entellan kullanılarak süreli preparatlarıyapıldı. Then the cellophanediscs with their fungus colony are carefulIy Iifted from the surface of the, medium and stained with Periodic acid Schiff method or analin blue. For the preparatianof permanent mounts theyare passed through graduated series of ethylalcohol and transfered ·to xYloL. Then theyare mounted: on microscope slides' İnCanada ba1sam. Eupara1 or Entellen. .'This technique perrnit the preparation of permanen-t môuuts r-etaıning diagnosticcharacteristics of spore stages and mycelium besides giving an opportunity ofobserving and photographing the development of a fungus on cultiıre mediums.
