ÖZET : Bu çalışmada, yerli Doğu Anadolu Kırmızısı melezi sığırlarında Prolaktin genine ait polimorfik bölgeler PCR-RFLP yöntemiyle incelenmiş ve bölgelere ait gen ve genotip frekansları belirlenmiştir. Analizde 5’UTR bölgesi için HaeIII, 4.ekzon bölgelesi için RsaI restriksiyon enzimleri kullanılmıştır. PCR-RFLP analizi sonucunda PRL geni 5’UTR bölgesinde populasyon monomorfik bulunmuştur. PRL geni 4.ekzon bölgesinde AA, AB ve BB genotip frekansları melez populasyonda sırasıyla 0.49, 0.44. ve 0.07 olarak tespit edilmiştir. Prolaktin geni 4. ekzon RsaI polimorfizm bölgesinde populasyonun Hardy-Weinberg dengesinde (p>0.05) olduğu görülmüştür. Anahtar Kelimeler: Prolaktin, Polimorfizm, DAK melezi, PCR-RFLP. Polymorphism of Prolactin Gene in the East Anatolian Red Crosbreds ABSTRACT : In this study, in the native East Anatolian Red Crosbreds, different polymorphic site of Prolactin gene was examined by PCR-RFLP method and the gene and genotype frequencies were determined. In the analysis of 5’UTR, we used HaeIII while RsaI restriction enzyme for exon 4. PCR-RFLP analysis was resulted with monomorphic genotype for 5’UTR in the population. AA, AB and BB genotype frequencies in exon 4 were 0.49, 0.44 and 0.07 for crosbreeds EAR.
