Farklı Bitki Büyüme Düzenleyicilerinin Türkiye'de Yetişen Cyclamen persicum Mill. ve Ticari "MelodyF1" Çeşidinde Ovül Kültürü Üzerine Etkileri

Bu çalışmada NAA ile BA ve Kinetinin yabani Cyclamen persicum Mill. ve ticari çeşit Melody F1"de ovül kültürü üzerine etkileri araştırılmıştır. Cyclamen persicum Mill. türü için etkili kallus oluşumu, 1.5 mgL-1 NAA +1.5 mgL-1 BA içeren ½ MS içeren besi yerinde %95 olarak ve 1.5 mgL-1 NAA + 1.5 mgL-1 Kinetin içeren ½ MS besi yerinde ise %81 olarak gözlemlenmiştir. Somatik embriyolar, sırasıyla 1.5 mgL-1 NAA + 1.5 mgL-1 BA (68%) ve 1.5 mgL-1 NAA + 1.5 mgL-1 Kinetin (59%) içeren ½ MS besi yerinde gözlemlenmiştir ve Cyclamen persicum Mill. için somatik embriyoların çimlenmesi hormonsuz MS besi yerinde %52 ve %57 olarak saptanmıştır. Melody F1 genotipinde kalluslar sırasıyla 1.5 mgL-1 NAA + 1.5 mgL-1 BA içeren ½ MS besi yerinde %95, 1.5 mgL-1 NAA + 1.5 mgL-1 Kinetin içeren besi yerinde %92 olarak gözlemlenmiştir. Somatik embriyolar aynı kallus oluşturma besi yerinde gözlemlenmiştir. Melody F1türünde ovül kültürü için embriyoların çimlenmesi hormon içermeyen MS besi yerinde %91 olarak belirlenmiştir. Aklimatizasyon oranı, C.persicum türü için %89, Melodi F1 %92 olarak belirlenmiştir. Bu çalışmada iki farklı Cyclamen genotipinde ovül kültürü için en iyi kallus oluşumu ve somatik embriyo besi yeri belirlenmiş ve etkili köklenme ve yüksek aklimatizasyon oranı gözlemlenmiştir

Effects of Different Plant Growth Regulators on Ovule Culture of Turkish Cyclamen persicum Mill. and Commercial Variety "Melody F1"

In this study, the effects of NAA (1-Naphthaleneacetic acid) with BA (6-Benzylaminopurine) and Kinetin on ovule culture in wild type Cyclamen persicum Mill. and commercial variety Melody F1 were investigated. Efficient callus initiation was observed on ½MS (half strength) medium including 1.5 mgL-1 NAA +1.5 mgL-1BA with response of 95% and 1.5 mgL-1 NAA + 1.5 mgL-1 Kinetin as 81 %. Somatic embryos (SE) were observed on MS medium containing 1.5 mgL-1 NAA + 1.5 mgL-1 BA (68%) and 1.5 mgL-1 NAA + 1.5 mgL-1Kinetin (59%) respectively and germination of SE was detected on plant growth regulator (PGR) free ½ MS medium for Cyclamen persicum Mill. as 52% and 57%. Callus was observed on ½ MS medium with 1.5 mgL-1 NAA + 1.5 mgL-1 BA and 1.5 mgL-1 NAA + 1.5 mgL-1 Kinetin with the response of %95, %92 respectively in Melody F1 genotype. Somatic embryos were obtained from the same callus induction medium (CIM). Germination of embryos was determined as 91% on PGR free MS medium for ovule culture in Melody F1. Acclimatization ratio was determined as 89% for C. persicum, %92 for Melody F1. Survival rates were % 68 for C. persicum and %74 for Melody F1. In the present study, best callus induction and somatic embryo medium were determined from the ovule explants for two Cyclamen genotype and efficient rooting and high acclimatization ratio were observed
