Anthurium sp.'de Tomato Spoted Wilt Virus (TSWV) Enfeksiyonu ve Moleküler Karakterizasyonu

Çalışma Antalya/Türkiye'de yetiştirilen Anthurium sp,'de (Anthurium scherzerianum) bitkilerinde virüs benzeri simptomlar görülmesi üzerine 2015 yılında gerçekleştirilmiştir. Gözlemler sırasında Flamingo çiçeği bitkilerinin yapraklarında, sistemik klorotik mozaik ve düzensiz sarımsı koyu kahverengi nekrotik lekeler gözlenmiştir. Tanılanma için yaprak DAS-ELISA tekniğine göre AMV (Alfalfa Mosaic Virus), ArMV (Arabis Mosaic Virus), CMV (Cucumber Mosaic Virus), Poty (Group Poty Virus), SLRSV (Strawberry Latent Ringspot Virus), TMV (Tobacco Mosaic Virus), TNV (Tobacco Necrosis Virus), ToMV (Tomato Mosaic Virus), ToRSV (Tomato Ringspot Virus), Tospogrup (Tospovirus), TRSV (Tobacco Ringspot Virus), TSWV (Tomato Spotted Wilt Virus), Impatiens necrotic spot virus (INSV),PVX (Potato Virus X), SqMV (Squash mosaic virus) ve ZYMV (Zucchini yellow mosaic virus) gibi süs bitkilerinde bulunma olasılığı bulunan virüsler için testlenmiştir. DAS-ELISA sonuçları pozitif örneklerden ekstrakte edilen toplam RNA'ların RTPCR'da kullanılmasıyla doğrulanmıştır. RT-PCR, sense TSWV L1F ve antisense primerlerine TSWV L2R göre düzenlenmiştir. RT-PCR sonucu kılıf proteinine ait 276 bp cDNA bantları %2'lik agaroz jelde gözlenmiştir. RT- PCR ürünlerinin direkt sekanslaması ve yapılan BLAST sonucunda biber TSWV izolatları ile %96-99 oranında benzerlik gösterdiği tespit edilmiştir. Anthurium bitkisinde TSWV enfeksiyonu serolojik ve moleküler olarak saptanmıştır. Bu çalışma yalnızca TSWV'nin Anthurium bitkisindeki Türkiye için ilk kaydı olup aynı zamanda etmenin türler arasında kontrolsüz yayılımının kanıtıdır.

Infection and Molecular Characterization of Tomato Spotted Wilt Virus (TSWV) on Anthurium sp.

This research wasconducted on Anthurium sp (Anthurium scherzerianum)observed the virus-like symptoms which was grown in Antalya in Turkey in 2015. During the observations, systemic mosaic and irregular yellowish dark brown necrotic spots were recorded on Flamingo flower plants. To identification, leaf samples were tested by DAS-ELISA using a specific polyclonal antiserum AMV (Alfalfa Mosaic Virus), ArMV (Arabis Mosaic Virus), CMV (Cucumber Mosaic Virus), Poty (Group Poty Virus), SLRSV (Strawberry Latent Ringspot Virus), TMV (Tobacco Mosaic Virus), TNV (Tobacco Necrosis Virus), ToMV (Tomato Mosaic Virus), ToRSV (Tomato Ringspot Virus), Tospo (Tospovirus), TRSV (Tobacco Ringspot Virus), TSWV (Tomato Spotted Wilt Virus), Impatiens necrotic spot virus (INSV), PVX (Potato Virus X), SqMV (Squash Mosaic Virus) and ZYMV (Zucchini Yellow Mosaic Virus) presence and identification of common ornamental plant viruses. Then samples which were positive in DAS-ELISA were confirmed by RT-PCR using total RNA extracted from leaves RT-PCR assay was conducted by primer pairs Sense TSWV L1r and Antisense TSWVL2f. As a result of RT-PCR, 276 bp cDNA bands were observed in 2% agarose gel. RT-PCR products were sequenced directly, and as the result of BLAST using datas from sequences, TSWV isolate in Anthurium showed 96-99 % similarity with different pepper isolates of TSWV. In Turkey, the occurrence of TSWV has reported on Anthurium sp. by serologically and molecular techniques. This study is not only the first record of TSWV in Turkey on Anthurium sp but also is evidence of uncontrolled distribution between plant species.
