Portakal kabuğu atıklarından üretilen biyokompozit ambalaj filminin aflatoksinlere karşı etkisinin incelenmesi

Bu çalışmada yenilikçi bir ürün olarak, portakal kabuğu atıklarından üretilen biyokompozit ambalaj filmine, aflatoksin önleyici özellik kazandırılması üzerinde durulmuştur. Biyokompozit filmin içeriğini portakal kabuklarından elde edilen pektin jeli, termoplastik nişasta ve kil oluşturmaktadır. Biyokompozit film, aflatoksinleri inhibe etmek üzere, enterik kaplamalarda kullanılan aktif bir ilaç malzemesi olan Selüloz Asetat Ftalat (CAP) ile daldırma yöntemiyle kaplanmıştır. Aflatoksin oluşumunun belirlenmesinde hedef gıda olarak kuru üzüm kullanılmış, sterilize edilen kuru üzüm örneklerinde toplam aflatoksin analizi (B1, B2, G1, G2) yapılarak, başlangıç aflatoksin miktarı (mg/g üzüm) belirlenmiştir. Aflatoksin konsantrasyonunun belirlenmesinde ekstraksiyon yöntemi kullanılmış ve analiz fluorometrik test cihazıyla yapılmıştır. Filmlerin kuru üzümde mevcut aflatoksinlere ve yeni aflatoksin oluşumuna etkisinin belirlenmesi için iki set deney yapılmış, yeni toksin oluşumunun belirlenebilmesi için de Aspergillus parasiticus kullanılmıştır. Altı kuyucuklumikroplate konulan kuru üzüm örneklerinin üzerleri biyokompozit film, CAP kaplanmış biyokompozit film ve karşılaştırma amaçlı olarak ticari ambalaj filmi ile kaplanmış ve 25 °C’de 5 gün süre ile inkübe edilmiştir. İnkübasyon sonrası kuru üzümlerde de aflatoksin analizi yapılarak başlangıç değerleriyle birlikte irdelenmiştir. Sonuçlara göre CAP ile kaplamanın biyokompozit ambalaj filminin aflatoksin ve üretimi üzerindeki inhibisyon etkisini arttırdığı belirlenmiş, CAP kaplı biyokompozit filmin yeni aflatoksin oluşumunun önlenmesi açısından ticari ambalaj filmine göre daha etkin olduğu saptanmıştır.

Effect of orange peels waste derived biocomposite film against aflatoxins

The aim of the study is to add an aflatoxin inhibition propertyto a novel biocomposite packaging film which is derived from orange peels waste and including thermo plastic starch and clay. Biocomposite film was coated with, Cellulose Acetate Phthalate (CAP), an enteric coating used as an active pharmaceutical ingredient, for aflatoxini nhibition. Total aflatoxin (B1, B2, G1, G2) analysis was realized on sterilized dry raisins. Before the test, all the dry raisins were sterilized and their initial total aflatoxin content (mg/g raisin) was determined. Then, a group of sterilized dry raisin were contaminated with Aspergillus parasiticus. Both sterilized and contaminated dry raisins were covered by the testing films and incubated at 25 °C forfivedays. After five days, final total aflatoxin contents in sterilized and contaminated dry raisin were measured to determine the existing aflatoxin and new aflatoxin growth, respectively. Total aflatoxin content was measured by a fluorometricmycotoxin test equipmentafterextraction. Resultsshowthat CAP has increased the inhibition effect of biocomposite film against existing aflatoxin and new synthesis. In addition to this, it was observed that CAP is more effective to new aflatoxin synthesis than commercial packaging film.
