Oksidanların ve Lipid Ekstraksiyonunun Buğday Proteinleri Üzerine Etkisinin Jel Filtrasyon Tekniği ile İncelenmesi

Bu çalışmada, Bezostaya çeşidi buğday unu, oksidan (askorbik asit veya potasyum bromat) katkı maddelerinin 50 ppm dozu ile op­timum yoğrularak hazırlanan hamurlar kullanılmıştır. Serbest ve bağlı lipidler petrol eteri ve suyla doyurulmuş n-bütanol ile ekstrakte edilerek uzaklaştırılmıştır. Lipid ekstraksiyonunun ve katkı maddelerinin un proteinleri üzerine etkisi jel filtrasyon tekniği kullanılarak araştırılmıştır. Ya­ğı alınmamış ve alınmış örneklerden AU (0.1 M asetik asit, 3M üre) çözücüsü ile ekstrakte edilen protein preparatlarının Sephacryl S-300 dol­gulu kolondan jel filtrasyon kromatografisi sonucunda üç pik elde edilmiştir. Oksidan maddeler proteinlerin kolonda alıkonma süresini değiştir­memiş, fakat proteinlerin relatif miktarlarının değişmesine sebep olmuştur. Bütün örneklerde lipid ekstraksiyonu, proteinlerin ekstrakte edilebi­lirliklerini ve agregatların moleküler ağırlığını azaltmıştır.

An Investigation on the Effects of Oxidants and Lipid Extraction on Wheat Proteins by Gel Filtration Technique (Turkish with English Abstract)

in this study, Bezostaya wheat flour and optimum mixed doughs prepared form this flour with oxidizing agents (ascorbic acid and potassium bromate at 50 ppm level) were used. Free and bound lipids were extracted with petroleum ether and water saturated n-buthanol, respectively. Effect of lipid extraction and additives on flour proteins were studied by gel filtration technique. Three peaks were obtained when proteins from defatted and undefatted samples were extracted with AU (0.1 M acetic acid, 3 M urea) solvent and separated using gel filtration chromatography on Sephacryl S-300. Oxidizing agents did not cause a change in the retention times of the proteins in the column, but caused some changes in their relative quantities. Lipid extraction caused decreases in the protein extractability and the molecular size of aggregates in all samples.
