Arthrospira platensis (Natron Gölü/Çad), Arthrospira maxima (Natron Gölü/Çad) ve Arthrospira platensis (Parachas Gölü/Peru)’in Suşlarının Büyüme Özelliklerinin Karşılaştırılması

Bu çalışmada Arthrospira platensis (Parachas Gölü/Peru), Arthrospira platensis (Natron Gölü/Chad) ve Arthrospira maxima (Natron Gölü/Chad)’nın yoğun üretimin ilk basamağı olan laboratuar koşullarındaki büyüme dinamikleri incelenmiştir. Deneme ortamı olarak saf suyla hazırlanan Zarrouk besin ortamı kullanılmıştır. Mikroalg türleri için ortam sıcaklığı 35 0C’ye, pH 9’a, aydınlatma 3000 lux ışık şiddetine ayarlanmış ve sürekli havalandırma yapılmıştır. Türlerin günlük olarak absorbansı spektrofotometre kullanılarak 680 nm’de ölçülmüş ve aynı zamanda Neubauer lamında hücre sayımları yapılmıştır. Arthrospira platensis’te (Natron ve Parachas suşu) 180 birim ile başlayan absorbans 14-15. güne kadar 1480 birime, A. maxima’da 290 birimle başlayan absorbans değeri 9-10. güne kadar 1190 birime ulaşmış sonraki günlerde azalma gözlenmiştir. Hücre sayıları da biyomasla orantılı olarak artış göstermiştir. Bu sonuçlar her üç türde de absorbans artarken hücre sayısının da arttığını ve aynı zamanda Arthrospira platensis’in A. maxima’ya göre daha sıcak ortamlarda gelişip üremesini sürdürebildiğini göstermiştir

Comparison of the Growth Characteristics of Arthrospira platensis (Natron Lake/Chad), Arthrospira maxima (Natron Lake/Chad) and Arthrospira platensis (Parachas Lake/Peru) Strains

This study explores the relation between the growth dynamics on the laboratory conditions which is the first step of mass production of Arthrospira platensis (Parachas Lake/Peru), Arthrospira platensis (Natron Lake/Chad) and Arthrospira maxima (Natron Lake/Chad). Zarrouk medium setting arranged in pure water has been used as experimental setting. The setting temperature for the species of microalgae is 35 ºC, pH is 9 and the lighting is fixed to the density of 3000 lux light and continual ventilation has been made. The daily absorbance of the species has been measured in 680 nm through a spektrophotometer, and cell counting has been made in Neubauer counting slide. The absorbance starting with 180 unit in Arthrospira platensis (Natron and Parachas water) as reached to the unit of 1480 on the 14th and 15th day, and the value of absorbance starting with 290 unit in A. maxima has reached the value of 1190 on the 9thand 10th day, and in the following days, decrease has been observed in these values. The number of cells has exhibited increases in line with biomass. These results have demonstrated in each three species that while the absorbance increases, the number of cells also increases and at the same time, Arthrospira platensis is capable of sustaining its growth and reproduction at hotter atmospheres when compared to A. maxima
